When you are facing the engine standing at the front of the car, #4 is the second cylinder from your left on front front bank of cylinders. See below:

1 3 5 7

2 4 6 8

Front

A fault coil pack can cause a mis-fire as well. Make sure #4 coil pack connection is good.

$375 sounds like a lot of money but with a 2000 it would not be out of the ordinary to neednew coil pack atthis time. Why not spend $75 first and get a mechanic torun the diagnosis to be sure.

does the engine actually misfire or is it just appearing in information center? It's just unclear by how it's written, and rather than being assumptive...?

Coil packs do fail, corrosion causes most of them. Unfortuneately one failing and the other failing 3-6 months later is not terriblely uncommon. They are somewhat expensive, you could go the used route "you take it off" types are best deals - 35.00 in my area.
